      目的 研究shRNA 靶向沉默CCDC132 基因对卵巢癌SKOV3 细胞增殖、迁移及侵袭的影响及其机制。方法 体外培养卵巢癌SKOV3 细胞株,设计CCDC132-siRNA 及空转序列,转染卵巢癌SKOV3 细胞,分别为sh-CCDC132 组及对照组,利用qRT-PCR检测各组细胞CCDC132 相对表达量,CCK8 法检测细胞增殖情况,划痕实验检测细胞迁移能力,Transwell 实验检测细胞侵袭能力,Western Blot 法检测Wnt/β-catenin 通路相关蛋白表达情况。结果 转染后,sh-CCDC132 组细胞中CCDC132 表达水平较对照组显著降低(P<0.01);sh-CCDC132 组细胞增殖率较对照组显著降低(P<0.01),迁移率下降(P<0.01)、侵袭细胞数显著降低(P<0.01),caspase-3、Bax 蛋白表达显著上调(P<0.01),而β-catenin(P<0.01)、Cyclin D1(P<0.01)和c-myc(P<0.05)蛋白表达水平显著降低。结论 沉默CCDC132 可抑制SKOV3 细胞增殖、迁移与侵袭能力,其具体机制可能与Wnt/β-catenin 信号通路有关。
英文摘要:
      Objective To study the effect of targeted silencing of the CCDC132 gene by shRNA on the proliferation,migration,and invasion of ovarian cancer SKOV3 cells and its mechanism.Methods Ovarian cancer SKOV3 cell line was cultured in vitro.CCDC132-siRNA and empty vector sequence were designed and transfected into ovarian cancer SKOV3 cells,which were the sh-CCDC132 group and the control group.The relative expression of CCDC132 in each group was detected by qRT-PCR.Cell proliferation was detected by CCK8 assay.Cell migration was detected by scratch assay.Cell invasion was detected by transwell assay,and Wnt/β-catenin pathway-related protein expression was detected by Western Blot.Results After transfection,the expression level of CCDC132 in the sh-CCDC132 group was significantly lower than that in the control group(P<0.01).The proliferation rate of the sh-CCDC132 group was significantly lower than that in the control group(P<0.01),the migration rate was decreased(P<0.01),and the number of inva sive cells was significantly decreased(P<0.01).The expression of caspase 3 and Bax protein was significantly up-regulated (P<0.01),while the expression levels of β-catenin (P<0.01),Cyclin D1(P<0.01) and c-myc(P<0.05) protein were significantly decreased.Conclusion Silencing CCDC132 can inhibit the proliferation,migration,and invasion of SKOV3 cells,and the specific mechanism may be related to the Wnt/β-catenin signaling pathway.
