目的 探讨HSP70 表达水平在启动正常分娩过程中的作用及机制研究。方法 将HSPA1A 和HSPA8 重组质粒及Si-HSPA8、Si-HSPA1A 转染WISH 细胞,采用ELISA 检测细胞中IL-8、FAS、BAX 和ESR2 表达水平,采用磷酸化抑制剂PD98059 处理WISH 细胞,qRT-PCR 和Western-blot 检测细胞中17 bβ-Estradiol、ERK1/2、p-ERK1/2、ERS2 和P-ERS2 表达。结果 转染48 h 后,转染pcDNA3.1 (-)-HSPA1A、pcDNA3.1 (-)-HSPA8 重组质粒组HSPA1A 和HSPA8 表达水平升高,SiRNA 干扰的HSPA1A 和HSPA8 表达水平降低;HSPA1A 组和HSPA8 组的BAX、FAS、ESR2 表达较NC 组降低,IL-8 表达较NC 组增加(P<0.05);与Si-NC 组比较,Si-HSPA1A 组和Si-HSPA8组的BAX、FAS、ESR2 表达较Si-NC 组增加,IL-8 表达Si-NC 组降低(P<0.05);与对照组比较,HSPA1A 和HSPA8过表达时17β-Estradiol 浓度升高,HSPA1A 和HSPA8 基因敲除后,17β-Estradiol 浓度与Si-NC 组比较,差异无统计学意义(P>0.04)。ERK1/2(PD98059)抑制剂处理WISH 细胞时,17β-Estradiol 的浓度较对照组显著升高(P<0.05);与对照组比较,HSPA1A 和HSPA8 过表达时细胞中的ERK1/2 和p-ERK1/2 显著降低,而HSPA1A 和HSPA8被敲低时细胞中的ERK1/2 和p-ERK1/2 较对照组显著升高。经PD 98059 处理的WISH 细胞中ESR2 和p-ESR2表达较对照组显著下降。结论 HSP70 可能通过ERK1/2 调控ESR2 的活化,进而可能刺激机体雌激素分泌以确保分娩过程顺利进行。 |

Objective To explore the role and mechanism of HSP70 expression level in initiating normal delivery.Methods HSPA1A and HSPA8 recombinant plasmids and Si-HSPA8,Si-HSPA1A were transfected into WISH cells.The expression levels of IL-8,FAS,BAX,and ESR2 in the cells were detected by ELISA.WISH cells were treated with phosphorylation inhibitor PD98059.The expression of 17 bβ-Estradiol,ERK1/2,p-ERK1/2,ERS2,and P-ERS2 in the cells was detected by qRT-PCR and Western-blot.Results 48 h after transfection,the HSPA1A and HSPA8 in the pcDNA3.1(-)-HSPA1A and pcDNA3.1(-)-HSPA8 recombinant plasmid groups increased,and the HSPA1A and HSPA8 induced by siRNA interference decreased.The expressions of BAX,FAS,ESR2 in the HSPA1A group,and HSPA8 group were lower than those in the NC group.The expression of IL-8 in the HSPA1A group and HSPA8 group was higher than that in the NC group (P<0.05).The BAX,FAS,ESR2 in the Si-HSPA1A,and Si-HSPA8 groups were higher than in the Si-NC group,and the expression of IL-8 was lower than in the Si-NC group (P<0.05).Compared with the control group,the 17β-Estradiol concentration increased when HSPA1A and HSPA8 were overexpressed.After the HSPA1A and HSPA8 genes were knocked out,there was no difference in 17β-Estradiol concentration compared with the Si-NC group(P>0.05).When ERK1/2(PD98059) inhibitor treated WISH cells,the 17β-Estradiol concentration was significantly higher than that of the control group (P<0.05).Compared with the control group,the ERK1/2 and p-ERK1/2 in the cells decreased significantly when HSPA1A and HSPA8 were overexpressed,while the levels of ERK1/2 and p-ERK1/2 in cells were significantly higher when HSPA1A and HSPA8 were knocked down.The expression of ESR2 and p-ESR2 in WISH cells treated with PD 98059 was significantly lower than that in the control group.Conclusion HSP70 may regμLate the activation of ESR2 through ERK1/2,which may stimμLate the body′s estrogen secretion to ensure a smooth delivery process. |
